Asporin (ASPN)
Asporin,a small leucine-rich repeat protein,encodes
an extracellular matrix protein that binds other
structural components of the extracellular matrix,including
collagen and TGF_.
Mice deficient in small
leucine-rich proteins,such as fibromodulin,or in biglycan
and fibromodulin,develop a mild and a severe
premature OA,respectively (38,39).Asporin mRNA is
also expressed abundantly in OA cartilage (37).Epidemiologic
studies in a Japanese population (40) have
shown that a specific aspartic acid repeat polymorphism
in ASPN is associated with inhibition of chondrogenesis
and increased susceptibility to OA (Table 3).In this
study,the D14 allele (referring to a unique repeat region
in the N-terminal region) was significantly more frequent
in patients with knee OA than control subjects,
while the D13 allele was significantly less common in
patients than in control subjects,suggesting a protective
effect.
