I am sorry for my previous behaivor.i depressed resently,Never thought the boyfriend of 2 years who is in American suddenly broke up with me,so I am sorry,but don't worry,After friend's encouraging and enlightenning me,now I am ok and I pluck up the courage to face it optimistically now.
I am writing to explain what I have done in the last 3 months.In the first month,I cultured HCT 116 cells and counted them with the help of CASY.Then I helped Sarah culture other cells and made sections in the other lab.In the second month,I practiced Western blot on PGK1 and Beta-actin.At the first time the line 3 had no trace for PGK1,I did it in the same way ,so i thought proteins might be not enough,i just used 100ug.I am not sure whether i have made a mistake detecting the content of protein.Maybe I need to measure protein twice and add more protein.It was not every good for Beta-actin,I did not know why,Sarah said it maybe owe to less protein,I did the R-T PCR as well and the result was ok.
In the last month,I continued to culture cells and do western blot and R-T PCR.I did western blot on beta-actin and another person did it on PGK1,we used the same quality of protein but different protein of cells,however the result on Beta-actin was bad.By which I think we need more protein to do western blot on Beta-actin.I made a lot of sections of tumors or nomal tissues.I did R-T PCR again although the last sample is not very good for we didn't have enough solutions as we needed in our lab.
In addition,I am continuing study english in my spare time and planning to do immnohistochemical experiments after the vacation.I would very much like to thank you for your help and everything you have done for me is greatly appreciated.
